Bromoacetyl[methyl-3H]choline is a highly specific label for the reduced acetylcholine binding site on the acetylcholine receptor from Torpedo californica. Only one of two binding sites per receptor monomer is susceptible to labeling. The labeled site is on the α chain of the receptor.  相似文献

Loss of tritium from specific positions in [3H,14C] aromatic hydrocarbons can elucidate their binding site(s) to DNA and RNA and indicate the mechanism of activation. Studies of tritium loss from [6-3H,14C]benzo[a]pyrene(B[a]P), [1,3-3H,14C]B[a]P, [1,3,6-3H,14C]B[a]P, [6,7-3H,14C]B[a]P, and [7-3H,14C]B[a]P were conducted in vitro using liver nuclei and microsomes from 3-methylcholanthrene-induced Sprague-Dawley rats and in vivo on the skin of Charles River CD-1 mice. The relative loss of tritium from [3H, 14C]B[a]P was measured after binding to skin DNA and RNA, to nuclear DNA, and to native and denatured calf thymus and rat liver DNA's and poly(G) by microsomal activation. In skin, nuclei, and microsomes plus native DNA, virtually all B[a]P binding occurred at positions 1,3 and 6; while with microsomes plus denatured DNA or poly(G), B[a]P showed no binding at the 6 position and a small amount at the 1 and 3 positions. In vivo and with nuclei, binding at the 6 position predominated. Little loss of tritium from the 7 position was seen; this was expected because binding at this position is not thought to occur. This confirms the interpretation of loss of tritium as an indication of binding at a given position. These results demonstrate that the use of microsomes to activate B[a]P is not a valid model system for delineating the in vivo mechanism of B[a]P activation, and support previous evidence for one-electron oxidation as the mechanism of activation of hydrocarbons in binding to nucleic acids.  相似文献

Summary A study was made of the morphology of the adenohypophysis in immature harp seals and the fine structure of cellular components of the pars tuberalis, pars intermedia and pars distalis was described.The pars intermedia composed 8–15% of the hypophysis and contained colloid filled vesicles similar to those found in the other mammalian species.The pars distalis cells were grouped into more or less well defined regions, thus facilitating the correlation of cellular identification from both light and electron micrographs. Five chromophilic cell types were tentatively identified, one acidophil (putative somatotroph), four basophils (3 putative gonadotrophs and one cell type with the characteristics of both corticotrophs and thyrotrophs) and non-granulated stellate cells. The absence of a positive prolactin cell identification was thought to be due to the immaturity of the seals used in the study.The mercury exposure experiment was supported by a contract grant from the Halifax Laboratory of Fisheries and Marine Service. Drs. Uthe and Freeman of that laboratory also carried out the methyl mercury analyses. We recognize the support in maintaining the seals provided by Mrs. C. Rae, Mrs. H. Pedersen, Mr. S. A nuclear-polyhedrosis virus (NPV) of the silkworm, Bombyx mori, which forms an icosahedral inclusion body, was transmitted to larvae of the rice stem borer, Chilo suppressalis. Serial passages of Bombyx NPV in the alternate host by injecting the supernatant of diseased hemolymph produced inclusion bodies with cuboidal and other shapes that differed from the original shape formed in Bombyx. These different shapes increased with times of passages, and after the twelfth passage, only cuboidal inclusion bodies were formed. The icosahedral inclusion bodies in B. mori and the cuboidal inclusion bodies in C. suppressalis occluded singly enveloped virions of the same size (350 × 75 nm), but the cuboidal inclusion bodies contained only a few virions and a large number of membraneous spherical structures. The formation process of the cuboidal inclusion body differed from that of the icosahedral. At first, irregularly branched inclusion bodies containing “vacant” spaces appeared in the infected nuclei. The bodies grew larger with the deposition of protein in the spaces between the branches, and this was accompanied with the occlusion of a large number of membraneous structures formed in the vicinity of the inclusion bodies, which became cuboidal in shape.  相似文献

Summary Scanning and transmission electron microscopy were used to study the inner architecture of the frog lung. In some specimens the alveolar surface mucus layer was removed to permit the examination of underlying features. The inner surface of the frog's lung is covered by a layer of microvilli belonging to only one type of epithelial cells. The boundaries of these epithelial cells are demarcated by small ridges. Different degrees of lung expansion cause variations of the surface topography. The morphology of certain surface features is examined in detail. Several methods of drying the specimens are compared.The author wishes to thank Dr. I. E. Abstract

In a lysimeter study with young beech trees, the effects of elevated ozone concentration on the decomposition and fate of nitrogen in 15N‐labeled leaf litter were analyzed after one growing season. Nitrogen in the litter was dominated by a relatively inert, residual fraction, but easily decomposable nitrogen was present in substantial amounts. Nitrogen loss was significantly higher at twice‐ambient ozone which was largely attributed to an enhanced mobilization of residual nitrogen. Enhanced mobilization of nitrogen from litter at twice‐ambient ozone exposure resulted in additional 15N incorporation into the soil down to 30 cm depth. Only 0.41–0.62% of the nitrogen in the litter was incorporated into plant material at both ozone concentrations. Twice‐ambient ozone exposure changed the distribution of the nitrogen taken up from litter inside the beech trees in favor of the shoot, where it may have been used in biosynthetic processes required for defense reactions.  相似文献

Twenty-five study sites were established along a 57-km-long transect in order to estimate the impact of an oil refinery, mainly emitting sulphur dioxide (24000 t yr−1), on forest soil (F/H-horizon) chemistry and microbiology. The study demonstrated the existence of a pollution gradient which was best represented by the logarithm of the concentration of vanadium in the analyzed F/H soil layer. Of the soil microbial characteristics measured, including length of fungal hyphae, soil respiration, microbial biomass C and N, and percentage mass loss of Scots pine (Pinus sylvestris) needle litter, only fungal hyphal length was suppressed by the pollution load. No reduction in basic cations (Ca, Mg, K, and Na) in the F/H-horizon, or enrichment of soluble aluminum in the F/H-horizon of the Scots pine forest could be detected to result from the deposition.  相似文献

The pseudo-capacitive behaviour of a high surface area carbon veil electrode in a tubular microbial fuel cell (MFC) was investigated as a mechanism to enhance power quality and energy efficiency. Accumulated charge and energy from the anodic biofilm after prolonged open circuit times (1–120 min) were compared against equivalent periods of steady state loading (R = 100–3000 Ω). A significant difference in the amount of accumulated charge with different loads was observed, resulting in 1.051 C (R = 100 Ω) compared to 0.006 C (R = 3 kΩ). The automated application of short open and closed circuit (0.5–10 s) cycles resulted in an increase of power/current production (closed circuit alone), but presented lower efficiency considering entire open and closed period. The cumulative charge on the carbon veil electrode with biofilm was 39,807 C m−2 at 100 Ω. Electrochemical Impedance Spectroscopy (EIS) showed that the Helmholtz layer presented a double layer capacitance of more than ten times the biofilm on electrode. The results indicate that the capacitive behaviour could be utilized to increase the power quality, i.e. its availability/applicability with respect to the operation of low power consuming devices.  相似文献

干旱区植物叶片形态可塑性是植物适应高温干旱环境的重要生存策略, 但目前仍缺乏直观的数据予以证明。该研究应用热成像技术和图像分析技术, 同步测定真实叶片与模拟叶片的叶温、形态及风速、辐射和温度等环境参数。研究结果显示: 在干旱、高温环境下, 除了蒸腾, 叶片形态变化也是调控叶温的重要因子。干旱区植物叶片变小, 有利于加速叶片与环境的物质及热量交换, 从而达到降低叶温的目的。样地数据显示, 在高温、低风速环境下, 叶片宽度每减少1 cm, 叶片表面温度降低约2.1 ℃, 而模拟叶片叶宽度每减少1 cm, 叶片表面温度降低0.60-0.86 ℃。该研究对深入理解植物生存策略与环境适能力具有重要意义。  相似文献
